目的观察亚低温条件下离体大鼠肝细胞悬浮培养后的完整性和功能。方法离体肝细胞悬浮于添加有葡萄糖和牛血清白蛋白(BSA)的碳酸氢盐缓冲介质内30℃下孵育48 h,期间及其后测定细胞的完整性和功能指标。结果30℃条件下离体0、24及48 h内白蛋白合成及Ⅰ相药物生物转化能力差异无统计学意义(P>0.05),流式细胞仪活细胞计数和台盼蓝清除率显示细胞完整性差异无统计学意义(P>0.05)。结论肝细胞混悬液内的离体肝细胞在亚低温条件下能够保持较高活力,为制备应用于生物人工肝的离体肝细胞混悬液提供了可行性。
Objective To observe the integrity and function of isolated rat hepatocytes suspension culture under mild hypothermia. Methods The isolated hepatocytes were suspended in bicarbonate buffer medium supplemented with glucose and bovine serum albumin (BSA) for 48 h at 30 ℃. The integrity and function of the cells were measured during and after the incubation. Results There was no significant difference in albumin synthesis and phase Ⅰ drug biocompatibility between 0, 24 and 48 h in vitro at 30 ℃ (P> 0.05). Flow cytometry showed that viable cell count and trypan blue clearance rate showed that cells There was no significant difference in completeness (P> 0.05). Conclusion The isolated hepatocytes in hepatocyte suspension can maintain high viability under mild hypothermia, which is feasible for preparation of isolated hepatocyte suspension for bioartificial liver.
