目的检测35例肾癌标本及配对正常组织标本中miR-10b的表达,分析miR-10b表达水平与患者临床特征之间的关系,为miR-10b后续功能研究奠定基础。方法提取35例手术切除的肾癌及癌旁正常组织标本中的总RNA,反转录获得cDNA,实时荧光定量PCR(qRT-PCR)检测miR-10b的表达水平,统计分析其表达水平与患者临床病理特征之间的关系。结果 35例组织标本中26例(74.29%)肾癌组织miR-10b表达水平下降(P<0.05),且下降和患者年龄、性别、肿瘤分期分级等无明显相关(P>0.05)。结论 miR-10b在肾癌组织中表达明显下调,提示其与肾癌发生有一定关系。
Objective To detect the expression of miR-10b in 35 specimens of renal cell carcinoma and paired normal tissues, and to analyze the relationship between the expression of miR-10b and the clinical features of the patients, so as to lay a foundation for further study of miR-10b. Methods Total RNA was extracted from 35 surgically resected neoplasms and adjacent normal tissues. CDNA was reverse transcribed and the expression of miR-10b was detected by real-time quantitative PCR (qRT-PCR). The expression levels of miR-10b were analyzed statistically The relationship between clinicopathological features. Results The expression of miR-10b in 26 cases (74.29%) of the 35 renal tissues was significantly lower than that of the normal controls (P <0.05). There was no significant correlation between the decline and the age, gender and tumor stage (P> 0.05). Conclusion The expression of miR-10b in renal cell carcinoma was significantly down-regulated, suggesting that it may be related to the occurrence of renal cell carcinoma.
