目的 :探索丙型肝炎病毒 (HCV)体外感染成年树肝细胞形态学的变化。方法 :HCVRNA阳性血清与培养肝细胞共同孵育后 ,收集不同时相标本并用逆转录聚合酶链反应 (RT -PCR)检测细胞内和上清中正负链RNA ,光镜、电镜观察感染细胞形态学变化。结果 :光镜下未发现感染细胞形态学变化 ,电镜下发现胞浆内含有大量的脂肪空泡、多泡体 ,并可见疑似病毒样结构。结论 :丙型肝炎病毒体外不仅能感染树肝细胞 ,而且能直接致肝细胞病变
Objective: To explore the morphological changes of adult hepatocytes infected with hepatitis C virus (HCV) in vitro. Methods: After incubated with cultured hepatocytes, HCV RNA positive sera were harvested and collected at different time points. Positive and negative RNAs were detected by reverse transcriptase-polymerase chain reaction (RT-PCR). The morphological changes of infected cells were observed under light and electron microscopes Learning changes. Results: Morphological changes of infected cells were not observed under light microscope. The cytoplasm was found to contain a large number of fat vacuoles and multivessels under electron microscopy, and the virus-like structure was observed. Conclusion: Hepatitis C virus can infect not only hepatocytes in vitro, but also hepatocellular disease
