以野生鹿药(Smilacina japonica A.Gray)的根茎及根为对象,研究了鹿药多糖的超声提取工艺并测定了多糖含量。结果表明:水为提取剂,超声提取的最佳工艺参数为料液比1∶20(g∶mL),温度30℃,超声时间20 min。多糖含量测定采用苯酚-硫酸法,葡萄糖为对照品,最大吸收波长490 nm,回归方程为A=13.049C+0.003 7,r=0.999 8,该方法的精密度、重复性、加样回收率、稳定性的RSD分别为3.80%、4.43%、0.04%、0.16%。采用最佳工艺提取和含量测定,测得鹿药中多糖含量为(23.79±0.02)%。
The rhizomes and roots of wild Deer (Smilacina japonica A. Gray) were studied. The ultrasonic extraction of deer polysaccharide was studied and the content of polysaccharide was determined. The results showed that the optimum extraction parameters were as follows: the ratio of solid to liquid was 1:20 (g: mL), the temperature was 30 ℃ and the ultrasonic time was 20 min. The polysaccharide content was determined by phenol-sulfuric acid method and glucose as reference substance. The maximum absorption wavelength was 490 nm. The regression equation was A = 13.049C + 0.003 7, r = 0.999 8. The precision, repeatability, The RSDs of stability were 3.80%, 4.43%, 0.04%, 0.16% respectively. Using the best extraction and determination of the content, the content of polysaccharide in deer medicine was (23.79 ± 0.02)%.
