[目的]探索魔芋芽鞘外植体愈伤组织的发生方式及不同处理条件对其愈伤诱导的效应程度,优化魔芋离体培养条件。[方法]以陕南地区魔芋芽鞘为试材,选用6-BA、NAA、KT、2,4-D 4种激素组合形成8种激素配比处理,统计不同处理条件下魔芋芽鞘愈伤组织的诱导情况。[结果]魔芋芽鞘外植体愈伤组织的发生表现出明显的多位点发生与多种不同形态;不同处理条件下魔芋芽鞘外植体脱分化启动与愈伤组织诱导效果存在显著差异(P<0.01)。[结论]当培养条件为MS+1.0mg/L 6-BA+2.0 mg/L NAA或MS+1.0 mg/L KT+2.0 mg/L 2,4-D时,魔芋芽鞘外植体脱分化诱导愈伤组织效果相对最佳。
[Objective] The research aimed to explore the way of callus formation and the effect of different treatment conditions on the callus induction of Konjak buds sheath explants, and to optimize the condition of in vitro culture. [Method] With 4 kinds of hormone combinations of 6-BA, NAA, KT and 2,4-D as the tested materials, the hormones of Konjac buds in southern Shaanxi were used as the experimental materials. Tissue induction. [Result] The occurrence of callus of konjak bud sheath explants showed obvious multi-locus occurrence and many different morphologies. Under different treatment conditions, the dedifferentiation of konjac bud sheath explants was significantly different from the callus induction effect (P <0.01). [Conclusion] The dediflora of Amorphophallus konjac bud explants were dedifferentiated when MS + 1.0 mg / L 6-BA + 2.0 mg / L NAA or MS + 1.0 mg / L KT + 2.0 mg / L 2,4-D Callus induction is relatively optimal.
