目的探讨金钗石斛多糖(NDP)对新生大鼠大脑皮质胶质细胞-神经元混合培养体系的作用。方法制备新生大鼠大脑皮质胶质细胞-神经元混合培养体系,分别用NDP与脂多糖(LPS)刺激并观察细胞生长情况,采用Western blot检测体系中炎症相关因子IL-1β、TNF-α、COX-2、IL-6的蛋白表达及相关调控信号p-NF-κB、p-p38、p-JNK、p-ERK的水平。结果LPS作用于大鼠胶质细胞-神经元混合培养体系时,炎症相关因子IL-1β、TNF-α、COX-2、IL-6的蛋白表达及相关调控信号pNF-κB、p-p38、p-JNK、p-ERK的水平明显升高。然而,NDP明显抑制了LPS诱导的系列变化。结论 NDP可阻遏LPS诱导的新生大鼠皮质胶质细胞-神经元混合培养体系中小胶质细胞和星形胶质细胞激活,从而保护神经元。
Objective To investigate the effect of dendrobium nobile polysaccharide (NDP) on glial cell-neuron mixed culture system in neonatal rats. Methods Neonatal rat glioma-neuronal mixed culture system was prepared. NDP and LPS were respectively used to stimulate and observe the cell growth. Western blot was used to detect the expression of IL-1β, TNF-α, COX-2, IL-6 protein expression and related regulatory signals p-NF-κB, p-p38, p-JNK, p-ERK levels. Results The expression of IL-1β, TNF-α, COX-2 and IL-6 in glioma-neuron mixed culture system induced by LPS and the related regulatory signal of pNF-κB and p- p-JNK, p-ERK levels were significantly increased. However, NDP significantly inhibited the series of changes induced by LPS. Conclusion NDP can protect neurons by inhibiting the activation of microglia and astrocytes in LPS-induced cortical glial cells-neuronal mixed culture system.
