用肾综合征出血热病毒(HFRSV)免疫Lou/c大鼠脾细胞与IR983F骨髓瘤细胞融合,获得11株能分泌特异性抗体的杂交瘤细胞系。用放射免疫沉淀、Westernblotting及免疫酶技术等对这一组大鼠单克隆抗体(单抗)进行了特性鉴定和病毒抗原性分析。发现11株单抗中,9株为抗核壳蛋白(NP)特异性,2株为抗G2糖蛋白。大多数抗NP单抗无中和活性及血凝抑制(HI)活性,但有2株例外,具有一定程度的中和或(和)HI活性。而抗G2单抗均只有一定程度的中和或(和)HI活性。在HFRSV抗原性分析方面,可初步将11株大鼠单抗分成组、亚组、野鼠型及亚型特异性单抗,并对上述单抗特性进行了讨论。
Spleen cells from Lou / c rat immunized with HFRSV were fused with IR983F myeloma cells to obtain 11 hybridoma cell lines secreting specific antibodies. The characteristics of this group of rat monoclonal antibodies (mAbs) and their antigenicity were analyzed by radioimmunoprecipitation, Westernblotting and immunoenzyme techniques. Nine of the 11 monoclonal antibodies were found to be specific for nucleocapsid protein (NP) and two to G2 glycoprotein. Most anti-NP mAbs had no neutralizing activity and hemagglutination inhibition (HI) activity, with 2 exceptions, with some degree of neutralization or (and) HI activity. The anti-G2 mAb only had a certain degree of neutralization or (and) HI activity. In the aspect of antigenicity analysis of HFRSV, 11 rat monoclonal antibodies were initially divided into groups, subgroups, wild-type and subtype specific monoclonal antibodies, and the characteristics of the above monoclonal antibodies were discussed.
