目的研究Cox7a2调控睾酮生成的影响及其机制。方法构建Cox7a2荧光表达载体,转染TM3小鼠睾丸Leydig细胞。ELISA法测定上清液中睾酮含量,Western blot检测类固醇合成快速调节蛋白StAR表达,荧光分光光度计检测ROS水平。结果 Cox7a2抑制LH诱导的睾酮合成,降低StAR蛋白的表达,升高ROS水平。结论 Cox7a2通过抑制类固醇合成快速调节蛋白StAR的表达调控睾酮分泌,至少和增高ROS水平有关。
Objective To study the effect of Cox7a2 on testosterone production and its mechanism. Methods Cox7a2 fluorescent expression vector was constructed and transfected into Leydig cells of TM3 mice. The content of testosterone in the supernatant was measured by ELISA, the expression of StAR was detected by Western blot, and the ROS level was detected by fluorescence spectrophotometer. Results Cox7a2 inhibited LH-induced testosterone synthesis, decreased StAR protein expression and increased ROS levels. Conclusion Cox7a2 regulates testosterone secretion by inhibiting the expression of StAR, a steroidogenic regulator of at least the increased ROS level.
