为探讨大豆转录因子GmWRI1在大豆种子油脂形成过程中的调控作用,本研究从大豆吉育72花后35d的种子中克隆得到GmWRI1全长cDNA序列,并对该基因编码的蛋白的理化性质、结构功能进行了初步分析;通过实时荧光定量PCR对花后不同时期籽粒中GmWRI1进行定量分析并对种子进行油脂含量的测定,推测GmWRI1的表达量可能与大豆籽粒中油脂含量有关。此外,利用重组DNA技术构建酵母表达载体pGBKT7-GmWRI1并转入AH109酵母菌株中,结果表明GmWRI1具有转录激活活性,属于转录激活子。
In order to investigate the regulatory role of soybean transcription factor GmWRI1 in soybean seed oil formation, the full-length cDNA of GmWRI1 was cloned from the seeds of Soybean Jiyu72 35 days after flowering. The physical and chemical properties, structure The quantitative analysis of GmWRI1 in seeds at different stages and the determination of oil content in seeds by real-time fluorescence quantitative PCR suggested that the expression level of GmWRI1 might be related to the oil content in soybean seeds.
