目的 研究人参皂苷 (GRS)对神经元分化细胞株PC12损伤及凋亡的影响。方法 采用神经细胞培养和流式细胞仪技术 ,以 β淀粉样蛋白 (Aβ)诱导PC12细胞损伤作为细胞模型 ,观察Aβ对PC12细胞的影响以及不同剂量GRS对PC12损伤的保护作用。结果 CRS 5 ,10 μg·ml-1明显减少Aβ介导PC12损伤时细胞内酶的释放 ,提高细胞的存活率。对Aβ诱导的细胞凋亡 ,GRS也有一定的拮抗作用。结论 人参皂苷能拮抗Aβ诱导的神经细胞凋亡 ,对Aβ引起的神经细胞毒性有一定的保护作用。
Objective To study the effects of ginsenosides (GRS) on the injury and apoptosis of neuronal differentiation cell line PC12. METHODS: Neuronal cell culture and flow cytometry were used to detect the injury of PC12 cells induced by β-amyloid protein (Aβ) as a cell model. The effect of Aβ on PC12 cells and the protective effect of different doses of GRS on PC12 injury were observed. Results CRS 5 and 10 μg·ml-1 significantly reduced the release of intracellular enzymes from Aβ-mediated PC12 injury and increased the cell survival rate. GRS also has some antagonism against Aβ-induced apoptosis. Conclusion Ginsenoside can antagonize the apoptosis induced by Aβ and has a protective effect on the neurotoxicity induced by Aβ.
