目的建立检测食品中氯霉素(CAP)的蛋白芯片技术。方法氯霉素是一种小分子半抗原,可与牛血清白蛋白偶联为CAP-BSA成为抗原。在醛基化玻片表面固定CAP抗体,将待测物CAP与一定量Cy3标记的CAP-BSA同时加到芯片上,采用竞争法的实验原理进行蛋白芯片的技术研究。结果氯霉素最低检测限为0.001μg/ml。结论本研究制备了氯霉素蛋白芯片,并能够实现检测高通量、灵敏快速、特异性强的目标。
Objective To establish a protein chip technology for the detection of chloramphenicol (CAP) in food. Methods Chloramphenicol is a small molecule hapten that can be coupled to bovine serum albumin as CAP-BSA to become an antigen. CAP antibody was immobilized on the surface of aldehyde-based glass slides, and CAP and a certain amount of Cy3-labeled CAP-BSA were simultaneously added to the chip. The protein chip technology was studied by the competition principle. Results The detection limit of chloramphenicol was 0.001μg / ml. Conclusion This study prepared chloramphenicol protein chip, and can achieve high-throughput detection, rapid and sensitive, high specificity of the target.
