建立了测定巨噬细胞(Mφ)等酸性磷酸酶(ACP)活力的简便而准确的新方法,以α萘酚磷酸氢钠为基质,用单偶联剂固红B盐作显色剂、测定酶水解产生的α萘酚。确定了小鼠腹腔Mφ、脾、胸腺样品的测定条件。硫羟乙酸盐刺激小鼠腹腔MφACP活性有显著性升高。环磷酰胺可使脾ACP活性呈先升高后下降的双相性变化。氢化泼尼松非常显著抑制脾ACP活性。讨论了影响测定的因素及药物作用的意义。
A simple and accurate new method for determining the activity of acid phosphatase (ACP) such as macrophage (Mφ) was established. Based on α-Naphtholate monobasic sodium phosphate, a single coupling agent, solid red B-salt, was used as a color-developing reagent. The alpha naphthol produced by enzymatic hydrolysis. The determination conditions of Mφ, spleen, and thymus samples in the abdominal cavity of mice were determined. The thiol acetate stimulated the increase of MφACP activity in the abdominal cavity of mice. Cyclophosphamide could increase the biphasic change of spleen ACP activity at first and then decreased. Prednisolone significantly inhibited spleen ACP activity. The factors that influence the determination and the significance of the drug effect are discussed.
