目的:观察益智宁对注意缺陷多动障碍(ADHD)动物模型自发性高血压大鼠(SHR)前额叶皮质、纹状体酪氨酸羟化酶(TH)、多巴胺转运体(DAT)蛋白和基因表达的影响。方法:将30只SHR随机分为3组,分别用益智宁煎剂、盐酸哌甲酯和生理盐水灌胃28天,采用免疫组化法和原位杂交法检测其前额叶皮质、纹状体TH、DAT蛋白和基因的表达。结果:免疫组化法和原位杂交法检测均发现:TH阳性率:益智宁组、盐酸哌甲酯组前额叶皮质、纹状体均升高,与生理盐水组比较,差异均有显著性意义(P<0.05),益智宁组升高较盐酸哌甲酯组更为明显(P<0.05);DAT阳性率:益智宁组、盐酸哌甲酯组前额叶皮质、纹状体均降低,与生理盐水组比较,差异均有显著性意义(P<0.05),益智宁组降低较盐酸哌甲酯组更为明显(P<0.05)。结论:益智宁治疗ADHD的机制可能与其影响调控中枢前额叶皮质-基底神经节环路的多巴胺神经信号传导相关。
OBJECTIVE: To observe the effect of Yizhining on prefrontal cortex, striatum tyrosine hydroxylase (TH), dopamine transporter (DAT) protein in spontaneously hypertensive rats (SHR) model of attention deficit hyperactivity disorder (ADHD) And the impact of gene expression. Methods: Thirty SHRs were randomly divided into three groups. The rats were treated with Yizhining decoction, methylphenidate hydrochloride and normal saline for 28 days respectively. Immunohistochemistry and in situ hybridization were used to detect the prefrontal cortex, Body TH, DAT protein and gene expression. Results: Immunohistochemistry and in situ hybridization showed that the positive rates of TH were significantly higher in the Yizhining and Methylphenidate groups than those in the normal saline group (P <0.05). The positive rate of DAT in YIZINGNING group was significantly higher than that in methylphenidate hydrochloride group (P <0.05). The positive rates of DAT in YIZINGNING group and methylphenidate group were lower than those in the prefrontal cortex and striatum (P <0.05). Compared with the methylphenidate hydrochloride group, the Yizhining Capsule group was significantly lower than the methylphenidate hydrochloride group (P <0.05). Conclusion: The mechanism of Yizhining in treating ADHD may be related to the influence of dopamine signal transduction on the central frontal cortex - basal ganglia circuit.
