目的 分析HLAI类抗原与肾透明细胞癌和卵巢癌的关联性 ,为确定与这些肿瘤的易感性和抵抗性有关的基因 ,并为寻找肿瘤特异性的抗原肽 ,开展肿瘤免疫诊断和治疗奠定基础。方法 实验采用微量淋巴细胞毒试验 ,分析 82例肾透明细胞癌和 2 1例卵巢癌患者外周血HLA A抗原和 B抗原的抗原频率。结果 与肾透明细胞癌和卵巢癌关联的HLAI类完全不同 ,其中与肾透明细胞癌关联的位点大大多于卵巢癌的位点 ,有A9、A11、A2 8、B15、B2 2、B2 7和B40 ;而与卵巢癌关联的HLAI类抗原位点只有A10、A36和B12 ,且卵巢癌关联位点的P值大于与肾透明细胞癌关联的HLAI类抗原的P值。经过校正P值计算后只有卵巢癌中的HLA A10抗原保持显著关联。结论 HLAI类抗原关联性研究显示 ,HLA A10可能为卵巢癌提供抵抗性。
OBJECTIVE: To analyze the association between HLA class I antigens and clear cell carcinoma of the kidney and ovarian cancer. In order to determine the genes related to the susceptibility and resistance of these tumors, and to find tumor-specific antigenic peptides and to establish the basis for tumor immune diagnosis and treatment . Methods Twenty-four patients with clear cell renal cell carcinoma and 21 patients with ovarian cancer were analyzed for the antigenicity of HLA-A and B antigen by using microcytotoxicity assay. The results were completely different from those in clear cell and ovarian cancers, with sites significantly associated with clear cell carcinoma of the kidney, with A9, A11, A2 8, B15, B2 2, and B2 7 And B40. The HLA class I antigen sites associated with ovarian cancer were only A10, A36 and B12, and the P value of ovarian cancer associated sites was greater than the P value of HLA class I antigens associated with renal clear cell carcinoma. Only the HLA A10 antigen in ovarian cancer remained significantly correlated with the corrected P value. Conclusion HLAI antigen association studies have shown that HLA A10 may provide resistance to ovarian cancer.
