目的:探讨内毒素对大鼠肝、肠组织内皮素受体(ETR)活性变化及内皮素A型受体mRNA转录表达的影响。方法:选用Wistar大鼠30只,分为对照组、内毒素组。自左股静脉给予10mg/kg内毒素,观察了伤后3、6、12、24h4个时相点肝、肠组织ETR活性和内皮素A型受体mRNA相对含量。结果:肝、肠组织ETR的解离常数无明显变化,最大结合数伤后3h明显降低,持续24h。内皮素A型受体mRNA相对含量均高于对照组。结论:内毒素能使肝、肠细胞膜表面ETR的最大结合数目降低;并在转录和翻译水平使内皮素A型受体合成增加。
Objective: To investigate the effect of endotoxin on the activity of endothelin receptor (ETR) and the transcription of endothelin A receptor mRNA in liver and intestine of rats. Methods: Thirty Wistar rats were randomly divided into control group and endotoxin group. 10 mg / kg endotoxin was given from the left femoral vein and ETR activity and relative content of endothelin A receptor mRNA in the liver and intestine tissues at 3, 6, 12 and 24 h after injury were observed. Results: The dissociation constants of ETR in liver and intestine tissues showed no significant change. The maximum binding number of ETR decreased significantly after 3 hours and lasted for 24 hours. Endothelin A receptor mRNA relative content were higher than the control group. Conclusion: Endotoxin can reduce the maximum number of ETR on the surface of liver and intestinal cell membrane, and increase the synthesis of endothelin A receptor at transcription and translation level.
