用高效液相色谱对摇瓶培养获得的19个虫草属菌株的菌丝体及胞外液中核苷类成分进行了分析比较。结果表明,菌丝体中含有的核苷种类较多,且不同菌株菌丝体核苷类成分含量差异显著,而胞外液中的核苷主要为虫草素。同时发现虫草素主要分泌到培养基中,胞外液虫草素的含量可比菌丝体中高出40倍。通过筛选,获得一株虫草素高产菌株,培养液中虫草素含量可达366mg/L。
Nineteen Cordyceps isolates from mycelia cultured in shake flask were analyzed by high performance liquid chromatography (HPLC) for their nucleosides in mycelium and extracellular fluid. The results showed that the mycelium contained more nucleosides, and the nucleosides content of mycelium in different strains was significantly different. The main nucleosides in extracellular fluid were cordycepin. At the same time, it was found that cordycepin was mainly secreted into the culture medium, and the content of extracellular cordycepin was 40 times higher than that of mycelium. Through screening, a strain of cordycepin-producing strain was obtained. The content of cordycepin in the culture solution reached 366 mg / L.
