目的通过研究Herbst矫治器引导成年大鼠下颌前伸后髁突软骨RUNX2蛋白表达水平的变化,探讨Herbst矫治器应用于成年患者的可行性。方法 60只14周龄雄性SD大鼠,根据实验周期(3、7、14、21和30 d)分为5组,每组内随机分为实验组(6只)和对照组(6只)。实验组大鼠全天佩戴Herbst矫治器引导下颌前伸,对照组大鼠不佩戴矫治器。实验后3、7、14、21和30 d分别处死各组大鼠,取右侧髁突组织,免疫组化检测髁突软骨中RUNX2蛋白的表达水平。结果随着观测时间的延长,对照组中大鼠髁突软骨RUNX2表达的IOD值呈现递减趋势,但各时间点差异并无统计学意义(P>0.05);除3 d组外,其余各时间点实验组大鼠髁突软骨RUNX2表达的IOD值均较对照组高(P<0.05),且在21 d时,其IOD值与对照组相差幅度最大。结论 Herbst矫治器能够诱导成年大鼠髁突发生适应性改建。
Objective To study the changes of RUNX2 protein expression in mandibular condyle of adult rats guided by Herbst appliance and to explore the feasibility of using Herbst appliance in adult patients. Methods Sixty male Sprague Dawley rats, aged 14 weeks, were divided into five groups according to the experimental period (3, 7, 14, 21 and 30 d), and each group was randomly divided into experimental group (6) and control group . The rats in the experimental group were given Herbst appliance to lead the mandibular anterior extension all day, while the rats in the control group did not wear the appliance. At 3, 7, 14, 21 and 30 d after the experiment, rats in each group were sacrificed, and the right condylar tissue was obtained. The expression of RUNX2 protein in condylar cartilage was detected by immunohistochemistry. Results With the prolongation of observation time, the IOD value of RUNX2 expression in rat condylar cartilage showed a decreasing trend, but there was no significant difference at each time point (P> 0.05). Except the 3 d group, The IOD values of RUNX2 in condylar cartilage of experiment group were higher than that of control group (P <0.05), and the difference between IOD value and control group was the largest at 21 d. Conclusions Herbst appliance can induce condylar reconstruction in adult rats.
