成模的18只GK大鼠随机分为3组(每组6只):林格式液对照组、空载体对照组、瘦素表达组。瘦素表达组分别通过尾静脉和胫骨前肌肉多点注射注入500ug表达质粒,对照组分别注入林格式液和空载体。观察各组大鼠血清瘦素、体重、腹膜后脂肪、血糖以及血脂的变化。结果:瘦素表达组的血清瘦素水平较两个对照组持续增加及体重、腹膜后脂肪以及血糖和血脂较对照组明显下降(P<0.01)。结论:GK大鼠体内瘦素水平增加和作用增强可以促进糖、脂代谢,提高胰岛素的敏感性,对其自发性2型糖尿病有积极、肯定的作用。
Eighteen GK rats were randomly divided into 3 groups (6 in each group): Lin format liquid control group, empty vector control group and leptin group. Leptin expression group were injected through the tail vein and tibialis anterior muscle injection of 500ug plasmid, the control group were injected Lin format fluid and empty vector. The changes of serum leptin, body weight, retroperitoneal fat, blood glucose and blood lipids in each group were observed. Results: Serum leptin levels in leptin-treated group were significantly higher than those in the two control groups (P <0.01). The levels of body weight, retroperitoneal fat, blood glucose and blood lipids in the leptin group were significantly lower than those in the control group (P <0.01). CONCLUSION: Increased leptin level and enhanced effect of GK can promote the metabolism of glucose and lipids, improve insulin sensitivity and have a positive and positive effect on spontaneous type 2 diabetes mellitus.
