【目的】探讨青钱柳叶水提物对阴虚内热型2型糖尿病大鼠中医表征和免疫功能的作用。【方法】将60只SPF级SD大鼠随机分为正常组,模型组,知柏地黄丸组,青钱柳叶水提物高、中、低剂量组。复制阴虚内热型2型糖尿病大鼠模型:大鼠给予高脂饲料喂养,喂养第4周时,按0.2 mg·kg~(-1)·d~(-1)剂量连续皮下注射甲状腺素(T4)7 d;第5周时,一次性腹腔注射链脲佐菌素(STZ)35 mg/kg,3周后造模结束。模型复制成功后,除正常组和模型组外,其余各组给予相应药物灌胃治疗4周。每周观察中医证候指标并评分。末次给药后,观察空腹血糖(FBG)值变化、血糖曲线下面积(AUC)及血清中尿素(Urea)含量;取肝脏称质量计算肝指数,并检测肝组织肝糖原含量;检测CD3~+、CD4~+和CD8~+T淋巴细胞百分比。【结果】与模型组比较,给予青钱柳叶水提物治疗之后,阴虚内热型糖尿病中医表征表现如背部温度上升、舌色鲜红、活动次数明显增加、爪色青紫及眼睛神采异常等均有明显的改善(P<0.05或P<0.01),FBG、AUC、Urea含量和肝脏系数下降(P<0.05或P<0.01),肝糖原含量升高,CD8~+T细胞百分比升高,CD4~+T细胞百分比及CD4~+/CD8~+值下降(P<0.05或P<0.01)。【结论】青钱柳叶水提物能改善大鼠阴虚内热型2型糖尿病中医表征及调节免疫功能。
【Objective】 To investigate the effect of extract of Cyperus rotundus on TCM characterization and immune function in type 2 diabetic rats with yin deficiency. 【Methods】 Sixty SPF SD rats were randomly divided into normal, model group, Zhibai Dihuang Wan group, high, medium and low dose of water extract of Cyperus L. The model of type 2 diabetes mellitus with fever of the febrile type was duplicated. The rats were fed with high fat diet, and the rats in the 4th week were injected subcutaneously with thyroxine (0.2 mg · kg -1 · d -1) T4) for 7 days. At the fifth week, STZ (35 mg / kg) was injected intraperitoneally once a week for three weeks. After the successful replication of the model, except for the normal group and the model group, the other groups were given the corresponding drugs for gavage for 4 weeks. Weekly observed TCM Syndrome Index and score. After the last administration, the changes of fasting blood glucose (FBG), the area under the curve of blood glucose (AUC) and the content of urea in the serum were observed; the hepatic index was calculated and the liver glycogen content was measured; +, CD4 ~ + and CD8 ~ + T lymphocyte percentage. [Results] Compared with the model group, after giving the extract of Cyperus Lolipopsis extract, the TCM manifestations of endogenous heat-type diabetes of yin deficiency syndrome such as increased back temperature, bright red tongue, increased activities times, abnormalities of claw cyanosis and abnormal eyes (P <0.05 or P <0.01). The content of FBG, AUC, Urea and hepatic coefficient decreased (P <0.05 or P <0.01), the content of hepatic glycogen and the percentage of CD8 + T cells increased, The percentage of CD4 ~ + T cells and CD4 ~ + / CD8 ~ + decreased (P <0.05 or P <0.01). 【Conclusion】 The extract of Cyperus rotundus can improve TCM symptoms and regulate immune function of type 2 diabetes with endogenous Yin deficiency in rats.
