为了用DNA 序列分析仪快速、大批量地对大片段DNA 进行序列分析研究,我们采用巢式定向渐进性截短缺失突变体构建方法,成功地对一株C型逆转录病毒基因的大片段DNA进行了缺失突变体的构建。实验结果显示,该方法效果好,在大片段DNA序列分析研究中具有推广应用价值。文内还对该方法的影响因素进行了讨论。
