目的:运用代谢组学方法考察不同配伍比例的天麻-川芎对偏头痛大鼠内源性物质代谢的影响,寻找潜在的生物标志物并分析其代谢途径,为传统中药药对的药理机制研究提供新方法。方法:将药材分成4组:天麻组(50 g)、川芎组(50g)、天麻-川芎A组(天麻-川芎为1∶4)、天麻-川芎B组(天麻-川芎为4∶1),各组药材分别用10倍量70%乙醇提取。采取注射硝酸甘油的方法建立大鼠偏头痛模型。结合PCA法与PLS-DA法分析不同配伍比例的天麻-川芎对偏头痛大鼠内源性物质影响,结合载荷图信息,寻找生物标志物。结果:代谢组学分析表明,与天麻-川芎B组相比,天麻-川芎A组样本点更接近正常对照组,而远离模型组;通过载荷图筛选出16个分类贡献率较大的变量,对其中的9个潜在生物标志物进行了结构鉴定和代谢途径分析。结论:代谢组学提供了反映机体整体状态的检测指标,天麻-川芎A组使偏头痛大鼠内源性代谢表型向正常大鼠回归,其作用机制可能涉及到氨基酸代谢及三羧酸循环等代谢过程。
OBJECTIVE: To investigate the effects of different compatibility proportions of Gastrodia elata and Chuanxiong on the metabolism of endogenous substances in migraine rats by means of metabonomics, search for potential biomarkers and analyze their metabolic pathways, and provide a theoretical basis for the study of pharmacological mechanism of traditional Chinese medicine new method. Methods: The herbs were divided into four groups: Tianma group (50 g), Chuanxiong group (50 g), Gastrodia elata and Chuanxiong group A (Gastrodia elata and Chuanxiong group 1: , Each group of herbs were extracted with 10 times the amount of 70% ethanol. The model of migraine was established by injecting nitroglycerin. Combined PCA and PLS-DA analysis of different compatibility ratio of Gastrodia elata and Chuanxiong on migraine rats endogenous substances, combined with the load map information, looking for biomarkers. Results: Metabolomics analysis showed that compared with the B group, Gastrodia elata - Chuanxiong group A sample point closer to the normal control group, and away from the model group; selected by the load map of the 16 categories of large contribution to the variable, Nine of the potential biomarkers were identified by structure and metabolic pathways. Conclusion: Metabolomics provides a measure of the overall status of the body. Gastrodia elata and Chuanxiong A group regulates the endogenous metabolic phenotype of migraine rats to normal rats, and its mechanism of action may involve amino acid metabolism and tricarboxylic acid cycle Other metabolic processes.
