目的:研究萱草花提取物在大鼠体内的血药浓度变化。方法:大鼠经口灌胃给药0.5g·kg-1·5m L-1,给药前后乙醚麻醉状态下,眼窝静脉窦采血120μL,分别在0、5、15、30、60、120、240、480和720min时间点采血,LC-MS/MS法测定血浆中芦丁含量。结果:大鼠灌胃给药萱草花提取物后,芦丁在30min达到最大血药浓度(Cmax)(13.50±6.38)ng/m L,其半衰期为(5.09±2.33)h。结论:芦丁可被迅速吸收,且相对于单体芦丁,萱草花提取物疗效持续时间较长。
Objective: To study the change of blood concentration of Hemerocallis fulva extract in rats. Methods: Rats were orally gavaged with 0.5 g · kg-1 · 5 m L-1, and 120 μL of orbital venous blood was collected before and after the administration of diethyl ether in anesthetized rats. The rats were randomly divided into 0, 5, 15, 30, 60, 120, Blood samples were taken at 240, 480 and 720 min, and plasma concentrations of rutin were determined by LC-MS / MS. Results: After administration of Hemerocallis fulva extract, rutin reached the maximum blood concentration (Cmax) (13.50 ± 6.38) ng / m L at 30 min and its half-life was (5.09 ± 2.33) h. Conclusion: Rutin can be rapidly absorbed, and compared with the monomer rutin, Hemerocallis fulva extract longer duration of efficacy.
