Epidemiological evidence shows that smoking is the main cause of lung cancer. However, although its promotion and/or combined carcinogenesis may be a determinant factor in the carcinogenic effects of tobacco, the mechanism by which smoking induces lung cancer remains unclear. This article not only reports a considerable amount of DNA single-stranded cleavage (SSB) in smoking-induced human cells, but also attributed this SSB to the “activated oxygen” produced by smoking. The authors used intriguing elution techniques to analyze unstable sites and SSB in DNA.
