目的:探讨外周血共刺激分子OX40和OX40L在系统性红斑狼疮(SLE)患者外周血T、B细胞上的表达及其意义。方法:采用流式细胞仪检测技术对62例SLE患者和40例健康志愿者外周血T细胞上OX40和B细胞上OX40L的表达水平进行分析。结果:与健康对照组比,SLE患者组外周血CD4~+T细胞OX40表达水平显著高于健康对照组,而CD8~+T细胞OX40表达水平较对照组无明显变化;SL正患者组外周血B细胞上OX40L表达率较健康对照组显著升高(P<0.05);SLE活动期患者组外周血CD4~+T细胞上OX40及B细胞上OX40L的表达率明显高于非活动组;SLE患者组治疗后OX40和OX40L表达率随病情好转而下降。结论:SLE患者外周血CD4~+T细胞上OX40及B细胞OX40L表达水平明显上调,随着病情的好转,CD4~+T细胞上OX40及B细胞OX40L表达水平明显下调,提示OX40/OX40L信号在T细胞与B细胞相互作用过程中可能有助于T细胞的持续活化,从而参与SLE的发生发展。
Objective: To investigate the expression and significance of peripheral blood costimulatory molecules OX40 and OX40L on peripheral blood T and B cells in patients with systemic lupus erythematosus (SLE). Methods: Flow cytometry was used to detect the expression of OX40L on OX40 and B cells in peripheral blood of 62 SLE patients and 40 healthy volunteers. Results: Compared with healthy control group, the expression level of OX40 in peripheral blood CD4 ~ + T cells in SLE patients was significantly higher than that in healthy controls, while the expression level of OX40 in CD8 ~ + T cells was no significant difference compared with healthy controls; The expression of OX40L in B cells was significantly higher than that in healthy controls (P <0.05). The expression of OX40 on CD4 ~ + T cells and OX40L on B cells in peripheral blood of SLE patients were significantly higher than those in inactive patients After treatment, the expression of OX40 and OX40L decreased with the improvement of their condition. Conclusion: The expression of OX40 and OX40L on CD4 ~ + T cells in peripheral blood of SLE patients was significantly up-regulated. With the improvement of disease, the expression of OX40L on OX40 and B cells on CD4 ~ + T cells was significantly down-regulated The interaction between T cells and B cells may contribute to the sustained activation of T cells, and thus participate in the development of SLE.
