目的探讨低强度脉冲式超声波(LIPUS)对体外培养大鼠软骨细胞增殖和细胞外基质合成的影响。方法将第2代SD大鼠关节软骨细胞随机分为对照组和实验组。实验组在培养过程中给予LIUPS刺激,强度30 mW/cm2,频率1.5 MHz,脉冲宽度200μs,脉冲重复频率1.0 kHz,作用20 min后细胞继续培养2 d。采用细胞计数试剂盒8检测软骨细胞的增殖,通过RT-PCR法检测软骨细胞中聚合素与Ⅱ型胶原的表达。结果与对照组相比,实验组细胞增殖明显增高(P<0.05),聚合素与Ⅱ型胶原的表达显著上调(P<0.05)。结论 LIPUS能够促进软骨细胞的增殖,增强Ⅱ型胶原、聚合素表达。
Objective To investigate the effect of low intensity pulsed ultrasound (LIPUS) on the proliferation of rat chondrocytes and the synthesis of extracellular matrix. Methods The second generation SD rat articular chondrocytes were randomly divided into control group and experimental group. The experimental group was given LIUPS stimulation at the intensity of 30 mW / cm2, the frequency was 1.5 MHz, the pulse width was 200 μs and the pulse repetition frequency was 1.0 kHz. The cells were cultured for another 2 days after the effect for 20 min. The proliferation of chondrocytes was detected by the cell counting kit 8, and the expression of type II collagen and collagen in chondrocytes was detected by RT-PCR. Results Compared with the control group, the proliferation of experimental group was significantly increased (P <0.05), and the expression of collagen and type Ⅱ collagen was significantly increased (P <0.05).
