文章采用传统的平板培养方法筛选能够高效降解人参自毒物质的微生物菌株,并通过16S核糖体DNA序列同源性分析结合显微形态特征鉴定降解菌株;通过液体培养结合GC-MS测定菌株生长情况及对自毒物质的降解效率,以明确其对自毒物质的降解特性。最终成功从六年生人参根围土中筛选出5株人参自毒物质高效降解细菌,液体培养状态下均能以人工添加的自毒物质为碳源生长繁殖。该研究结果表明,从人参根围土中筛选自毒物质降解细菌具有可行性,筛选的降解菌株可用于老参地中自毒物质的消除。
In this paper, the traditional plate culture method was used to screen microbial strains that can degrade ginseng self-toxic materials efficiently. The strains were identified by 16S ribosomal DNA sequence homology analysis and microscopic morphological characteristics. The growth of the strains was determined by liquid culture and GC-MS And the degradation efficiency of self-toxic substances to clarify its degradation characteristics of self-toxic substances. In the end, five strains of ginseng self-toxic materials were successfully screened from six-year-old ginseng root soils. Under the condition of liquid culture, artificial supplemented self-toxic substances could be used as carbon sources for growth and reproduction. The results showed that it is feasible to screen the self-toxic bacteria to degrade bacteria from the ginseng rhizosphere soil. The screened degrading bacteria can be used to eliminate the self-toxic substances in the ginseng root.
