目的 探讨大剂量甲基强的松龙 (MP)诱导大鼠胸腺淋巴细胞凋亡作用的存在。方法 采用流式细胞仪技术、琼脂糖凝胶电泳等方法对细胞凋亡状态进行检测。结果 大剂量MP注于大鼠体内后 ,可诱导大鼠胸腺淋巴细胞进入凋亡状态 ,注入后 6h ,无明显细胞凋亡 ,12h后出现凋亡改变 ,2 4h凋亡细胞改变明显增加 ,连续用药 2~ 3d ,虽凋亡细胞比率增加 ,但与用药 2 4h相比 ,差异无显著性。结论 甲基强的松龙体内注射后 ,以用药后 2 4h诱导胸腺淋巴细胞凋亡的作用为最强。
Objective To investigate the effect of high dose methylprednisolone (MP) on the apoptosis of thymus lymphocytes in rats. Methods Flow cytometry, agarose gel electrophoresis and other methods of apoptosis were detected. Results After injection of MP in rats, the thymus lymphocytes of rats were induced to apoptosis. Six hours after injection, there was no obvious apoptosis. After 12 hours, apoptotic changes were observed. The changes of apoptotic cells increased significantly at 24 hours Medication 2 ~ 3d, although the rate of apoptotic cells increased, but compared with the medication 24h, no significant difference. Conclusion Methylprednisolone has the strongest effect on inducing apoptosis of thymus lymphocytes 24 h after injection.
