测定液态发酵羊肚菌胞外多糖含量时,由于受到培养基中夹带的糖类物质(如淀粉、蔗糖)等物质的干扰,其有效羊肚菌胞外多糖的含量测定可能会受到较大影响。分别以苯酚-硫酸法和改进的ROBERTS铜方法进行羊肚菌胞外多糖测定。实验表明,用ROBERTS铜法所测多糖含量仅占苯酚硫酸法所测多糖含量的17.3%,证明用ROBERTS铜法测定羊肚菌胞外多糖更准确一些。
Determination of liquid fermentation Morchella extracellular polysaccharide content, due to the medium entrained carbohydrates (such as starch, sucrose) and other substances interfere with the determination of effective Morchella extracellular polysaccharide may be greatly affected . The Morchella esculenta extracellular polysaccharide was determined by phenol-sulfuric acid method and modified ROBERTS copper method respectively. Experiments show that the polysaccharide content measured by ROBERTS copper method only accounted for 17.3% of the polysaccharide content measured by the phenol sulfuric acid method, which proves that the determination of Morchella extracellular polysaccharide by ROBERTS copper method is more accurate.
