目的观察麻杏石甘汤对流感病毒感染小鼠肺部炎症与超微结构的影响。方法采用鼻腔接种法建立流感病毒小鼠肺部感染模型。实验设正常对照组、模型对照组、利巴韦林组和麻杏石甘汤组,经灌胃给药或给生理盐水7 d后,处理动物,常规方法检测肺重量指数;光学显微镜技术观察肺病理变化;透射电子显微镜技术观察肺组织超微结构。结果①与正常对照组比较,模型对照组小鼠肺指数明显升高(P<0.05),麻杏石甘汤组小鼠肺指数降低,与模型对照组比较,差异有统计学意义(P<0.05);②与正常对照组比较,模型对照组小鼠肺脏出现明显的病理变化,肺泡、细支气管等正常组织结构消失,肺泡内有大量炎症细胞浸润,麻杏石甘汤组小鼠肺组织病理变化显著改善,偶可见少量炎症细胞浸润;③与正常对照组比较,模型对照组小鼠肺组织超微结构受到破坏,核膜与质膜模糊不清,细胞浆内细胞器减少,出现空泡样病变,发现病毒颗粒,麻杏石甘汤组小鼠肺组织的细胞超微结构较完整,核膜与质膜清晰可见,细胞器比模型对照组丰富,细胞浆内空泡样病变程度明显低于模型对照组。结论麻杏石甘汤对A型流感病毒感染所致的肺部炎症与超微结构有改善作用。
Objective To observe the effect of Maxingshigan decoction on lung inflammation and ultrastructure in mice infected with influenza virus. Methods Nasal inoculation method was used to establish the lung infection model of influenza virus in mice. Results ① Compared with the normal control group, the lung index of the model control group was significantly increased (P <0.05), and the lung index of the mice in the Maxing Shigan Decoction group was lower than that of the model control group (P < 0.05) .②Compared with the normal control group, the pathological changes of the lungs in the model control group were obvious. The normal tissues such as alveoli and bronchioles disappeared. There was a large number of infiltration of inflammatory cells in the alveoli. The lung tissues of the mice in the Maxing Shigan Decoction group Pathological changes significantly improved, and even a small amount of inflammatory cells can be seen infiltration; ③Compared with the normal control group, the ultrastructure of the lung tissue of the model control group was destroyed, the nuclear membrane and plasma membrane were unclear, cytoplasm organelles decreased, vacuoles Like lesions and found that virus particles, Maxing Shigan group mice lung tissue ultrastructure is complete, the nuclear membrane and plasma membrane can be seen clearly, organelles than the model group is rich in cytoplasm vacuolar-like lesions was significantly lower In the model control group. Conclusion Maxingshigan Decoction can improve lung inflammation and ultrastructure induced by influenza A virus infection.
