目的 研究白血病抑制因子 (LIF)mRNA在早孕期小鼠子宫内膜的表达规律及LIF蛋白的组织定位。方法 用RT PCR半定量法测定LIFmRNA的表达 ;用免疫组化ABC法测定LIF蛋白的组织学定位。结果 LIFmRNA在早孕期小鼠子宫内膜呈时序性规律表达 ,孕早期升高 ,第 4天达到高峰 ,比未孕期明显增高 (P <0 .0 1 ) ,此后下降 ,到孕第 7天降到孕前水平 ;LIF蛋白主要定位在子宫内膜腺上皮和腔上皮 ,且第 4天染色最深 ,间质细胞无LIF表达。结论 LIF的表达高峰与小鼠孕卵着床时间吻合。因此 ,LIF可能参与了着床的启动 ,LIF参与着床可能是通过自分泌和旁分泌作用的结果。
Objective To investigate the expression of leukemia inhibitory factor (LIF) mRNA in mouse endometrium in early pregnancy and the localization of LIF protein. Methods The expression of LIF mRNA was detected by semi-quantitative RT-PCR. The histological localization of LIF protein was determined by immunohistochemical ABC method. Results The expression of LIF mRNA in the endometrium of mice during the first trimester was regularly and regularly increased in the first trimester of pregnancy, reaching the peak on the fourth day, significantly higher than that of the non-trimester (P <0.01), and then decreased to the seventh day of pregnancy To pre-pregnancy level. LIF protein mainly localized in the endometrial gland epithelium and luminal epithelium, with the deepest staining on day 4 and no expression of LIF on interstitial cells. Conclusion The peak expression of LIF coincides with the time of implantation of ovum in mice. Therefore, LIF may be involved in the initiation of implantation, and LIF involvement in implantation may be the result of autocrine and paracrine effects.
