直接观察体外培养人脐静脉内皮细胞(HUVEC)上清液中血管内皮舒张因子(EDRF/NO)的释放变化及对血小板聚集的抑制作用。发现:(1)缓激肽(BK)或细菌脂多糖(LPS)能刺激(EDRF/NO增加到原来的2倍,并且以孵育第1h增长最多。(2)L-精氨酸(L-Arginine)作为EDRF/NO合成原料,可以提高HUVEC上清液中EDRF/NO量,但是EDRF/NO合成酶抑制剂L-NG-nitro-arginine(L-NNA)与L-Arginine同时存在时则降低这个合成量。(3)经消炎痛处理过并含有EDRF/NO的HUVEC上清液对血小板有抑制作用、这种抑制作用由于L-NNA的存在而减弱。
To directly observe the release of vascular endothelial relaxing factor (EDRF / NO) in supernatants of human umbilical vein endothelial cells (HUVEC) in vitro and the inhibitory effect on platelet aggregation. The results showed that: (1) EDRF / NO increased by 2 folds and increased most at 1 hour after incubation with BK or LPS; (2) L-arginine (L- Arginine as an EDRF / NO synthetic material can increase the amount of EDRF / NO in the supernatant of HUVEC. However, the EDRF / NO synthase inhibitor L-NG-nitro-arginine (L-NNA) (3) HUVEC supernatants treated with indomethacin and containing EDRF / NO have an inhibitory effect on platelets, which is attenuated by the presence of L-NNA.
