目的:优选加味四君子汤的提取工艺。方法:以人参皂苷总提取量为指标,采用L9(34)正交试验考察乙醇体积分数及用量、提取次数、提取时间对醇提工艺的影响;以总多糖得率为指标,通过正交试验考察料液比、提取次数、提取时间对水提工艺的影响。采用HPLC测定人参皂苷含量,流动相乙腈-水(13∶87),检测波长203 nm;利用UV测定总多糖含量,检测波长489 nm。结果:最佳醇提工艺为加6倍量70%乙醇提取2次,每次1.5 h;人参皂苷提取量达7.69 mg·g-1。最佳水提工艺为加8倍量水提取3次,每次2 h;总多糖得率达2.18%。结论:优选的提取工艺稳定可行,可充分提取加味四君子汤中醇溶性和水溶性有效成分。
Objective: To optimize the extraction process of Sijunzi Decoction. Methods: The total amount of ginsenosides as an indicator, the use of L9 (34) orthogonal test of ethanol volume fraction and dosage, extraction times, extraction time on alcohol extraction process; total polysaccharide yield as an indicator, by orthogonal test The effects of feed ratio, extraction times and extraction time on water extraction process were investigated. The content of ginsenosides was determined by HPLC. The mobile phase was acetonitrile - water (13:87). The detection wavelength was 203 nm. The total polysaccharide content was determined by UV detection at 489 nm. Results: The optimum alcohol extraction process was extraction twice with 6 times 70% ethanol for 1.5 h each time. The extraction of ginsenosides reached 7.69 mg · g-1. The best water extraction process for 8 times the amount of water extracted three times, each 2 h; total polysaccharide yield of 2.18%. Conclusion: The optimal extraction process is stable and feasible, which can fully extract the alcohol-soluble and water-soluble active ingredients in Jiawei Sijunzi Decoction.
