目的:研究8-溴-7-甲氧基白杨素(8-bromo-7-methoxychrysin,BrMC)诱导人顺铂耐药卵巢癌A2780/DDP细胞凋亡作用及细胞凋亡过程中Akt蛋白磷酸化水平的变化。方法:用不同浓度的BrMC处理体外培养的A2780/DDP细胞,碘化丙啶(PI)染色流式细胞术(FCM)和细胞凋亡ELISA检测试剂盒检测凋亡性细胞死亡,Western Blot分析Akt蛋白磷酸化水平。结果:BrMC具有诱导A2780/DDP细胞凋亡作用,并且随药物浓度的增加而增强;同时BrMC以浓度依赖的方式下降Akt蛋白磷酸化水平。结论:BrMC诱导A2780/DDP细胞凋亡与其抑制Akt活化相关。
AIM: To investigate the apoptosis of human cisplatin-resistant ovarian cancer A2780 / DDP cells induced by 8-bromo-7-methoxychrysin (BrMC) and the phosphorylation of Akt protein during apoptosis Horizontal changes. Methods: A2780 / DDP cells cultured in vitro were treated with different concentrations of BrMC. Proliferation of apoptotic cells was detected by flow cytometry (PI) and flow cytometry (FCM), and Akt Protein phosphorylation levels. Results: BrMC induced the apoptosis of A2780 / DDP cells and increased with the increase of drug concentration. At the same time, BrMC decreased Akt protein phosphorylation in a concentration-dependent manner. Conclusion: Apoptosis induced by BrMC in A2780 / DDP cells is related to the inhibition of Akt activation.
