目的:研究经髂内动脉、耳缘静脉注射羟基喜树碱后,家兔血液及膀胱组织内部羟基喜树碱药物浓度的差异,以确定该药是否适合应用于经导管骼内动脉灌注治疗方案。材料与方法:(1)动物:A 组40只雌性白色家兔,分为动、静脉两大组,每大组内均按膀胱标本取样时间不同分为5小组。B 组16只雌性家兔,分为动、静脉两组,每组8只,进行动态血药浓度检测。(2)方法:动脉途径先将家兔耳缘静脉给药。药物剂量:1mg/kg,灌注时间为10分钟。(3)组织中药物浓度的测量:采用反相高效液相色谱法。结果:在分别经髂内动脉,耳缘静脉给药后,5、15、30、60、120分钟后膀胱组织中羟基喜树碱药物平均浓度明显差异,动脉途径分别为静脉途径的2.17、3.41、1.51、1.52、2.33倍。血液中平均药物浓度静脉途径分别为动脉灌注方法的1、92、2.03、1.77、1.92、1.40倍。结论:羟基喜树碱具备经导管动脉灌注的基本条件,可以应用于动脉化疗的方案中。
OBJECTIVE: To study the differences in the concentration of hydroxycamptothecin in blood and bladder tissue of rabbits after injecting hydroxycamptothecin through the internal iliac artery and the ear vein to determine whether the medicine is suitable for the treatment of endo-arterial infusion by catheter . MATERIALS AND METHODS: (1) Animals: A group of 40 female white rabbits were divided into two groups: arterial and venous. Each group was divided into 5 groups according to different sampling time of bladder specimens. B group of 16 female rabbits, divided into two groups of animals, venous, 8 in each group, for dynamic blood concentration test. (2) Method: The arterial route first rabbits ear vein administration. Drug dose: 1mg / kg, perfusion time of 10 minutes. (3) measurement of tissue drug concentration: using reversed-phase high performance liquid chromatography. Results: The mean concentrations of hydroxycamptothecin drugs in bladder tissue were significantly different at 5, 15, 30, 60 and 120 minutes after intravenous administration via the internal iliac artery and the marginal ear. The arterial routes were 2.17 and 3.41 , 1.51, 1.52, 2.33 times. The average blood concentration of drug intravenous route were arterial perfusion method 1,92,2.03,1.77,1.92,1.40 times. Conclusion: Hydroxycamptothecin has the basic conditions of transcatheter arterial infusion and can be applied in the scheme of arterial chemotherapy.
