背景与目的人参是我国传统中草药,其有效化学成分为人参皂甙,20(R)-人参皂甙Rg3抗肿瘤作用最显著,可以通过多种作用机制抑制肿瘤的生长和转移。肿瘤的发生和发展是一个极其复杂的过程,有多种蛋白因子参与,并涉及多种分子机制。人参皂甙Rg3抗肿瘤机制的研究为目前中药抗肿瘤研究热点之一。本研究的目的在于探讨Rg3上调乳腺癌细胞的FAS蛋白从而促进肿瘤细胞的凋亡。材料与方法四甲基偶氮唑蓝法检测20(R)-人参皂甙Rg3能抑制乳腺癌细胞MCF-7的增殖活力,并得出半数抑制浓度(IC50)。流式细胞仪检测20(R)-人参皂甙Rg3能诱导细胞凋亡和调节细胞周期。免疫细胞化学检测MCF-7细胞FAS蛋白表达。结果Rg3对MCF-7抑制作用随着浓度的增加而增加,IC50为200μg/ml,流式细胞仪检测Rg3使MAF-7的S期细胞比率明显增加,凋亡率明显增加。免疫细胞化学显示Rg3能使MCF-7的FAS蛋白表达增加。结论人参皂甙Rg3上调FAS表达诱导乳腺癌MCF-7细胞的凋亡。
BACKGROUND & OBJECTIVE Ginseng is a traditional Chinese herbal medicine in our country. Its effective chemical composition is ginsenosides. The anti-tumor effect of 20 (R) -ginsenoside Rg3 is the most significant. It can inhibit tumor growth and metastasis through a variety of mechanisms. Tumor occurrence and development is an extremely complex process, a variety of protein factors involved, and involves a variety of molecular mechanisms. Research on anti-tumor mechanism of ginsenoside Rg3 is one of the hot topics in antitumor research of traditional Chinese medicine. The purpose of this study is to investigate the role of Rg3 in upregulating FAS protein in breast cancer cells to promote tumor cell apoptosis. MATERIALS AND METHODS The proliferation of breast cancer cells MCF-7 was inhibited by 20 (R) -ginsenoside Rg3 by MTT method and the IC 50 was obtained. Flow cytometry detected 20 (R) - ginsenoside Rg3 can induce apoptosis and regulate cell cycle. Immunocytochemistry was used to detect FAS protein expression in MCF-7 cells. Results The inhibitory effect of Rg3 on MCF-7 increased with the increase of IC50 of 200μg / ml. The detection of Rg3 by flow cytometry increased the percentage of S phase cells and the apoptosis rate of MCF-7 cells. Immunocytochemistry showed that Rg3 increased the expression of FAS protein of MCF-7. Conclusion Ginsenoside Rg3 up-regulates FAS expression and induces apoptosis in breast cancer MCF-7 cells.
