目的观察胚胎Wistar大鼠心脏发育过程中转化生长因子(TGF)-β1、Smad3的表达,探讨其在胚胎心脏发育的意义。方法 28只月龄、体质量均相当的Wistar大鼠,随机分为7组,每组4只,分别在妊娠E12、E13、E14、E15、E16、E17、E18时麻醉,行剖宫术取出胎鼠,取其心脏,液氮保存。采用RT-PCR法检测其TGF-β1、Smad3在妊娠E12~E18胚胎心脏中的表达。结果妊娠E12~E18时,TGF-β1和Smad3表达由弱到强再减弱;TGF-β1表达高峰出现在E14,Smad3高峰出现在E17。结论 TGF-β1/Smad3信号通路对大鼠胚胎的心脏发育起一定的调控作用。
Objective To observe the expression of transforming growth factor (TGF) -β1 and Smad3 during embryonic heart development in Wistar rats, and to explore its significance in embryonic heart development. Methods 28-month-old Wistar rats of equal body weight were randomly divided into 7 groups with 4 rats in each group. They were anesthetized at E12, E13, E14, E15, E16, E17 and E18, respectively. Fetal rats, whichever is the heart, liquid nitrogen preservation. The expression of TGF-β1 and Smad3 in embryonic E12-E18 embryonic heart was detected by RT-PCR. Results The expression of TGF-β1 and Smad3 decreased from weak to strong at E12 ~ E18, the peak at TGF-β1 at E14 and the peak at Smad3 at E17. Conclusion The TGF-β1 / Smad3 signaling pathway plays a role in the regulation of cardiac development in rat embryos.
