获得兔骨保护素(OPG)基因并分析序列。从兔肱骨中提取总RNA,逆转录形成cDNA,用5′RACE策略扩增OPG基因,经琼脂糖凝胶电泳鉴定,并测序及进行序列分析。兔OPG基因全长1 540bp,编码400个氨基酸,与人OPG氨基酸序列相比,同源性为89%,而与大鼠等其它动物的同源性则在85%左右。5′RACE法成功获得了兔OPG基因的真实序列,为OPG基因的功能研究奠定了良好基础。
Rabbit osteoprotegerin (OPG) gene was obtained and analyzed for sequence. Total RNA was extracted from the humerus of rabbits and cDNA was reverse transcribed. The OPG gene was amplified by 5’RACE strategy, identified by agarose gel electrophoresis, sequenced and sequenced. The rabbit OPG gene is 1 540 bp in length and encodes 400 amino acids. Compared with the human OPG amino acid sequence, the rabbit OPG gene has a homology of 89%, while its homology with other animals such as rats is about 85%. The 5’RACE method successfully obtained the real sequence of rabbit OPG gene, which laid a good foundation for the study of the function of OPG gene.
