目的建立HPLC法,用于测定患者体内血浆中胺碘酮及其代谢产物去乙基胺碘酮血药浓度。方法采用色谱柱Ultimate C18(250 mm×4.6 mm,5μm)柱;流动相:醋酸-三乙胺缓冲溶液∶甲醇=19∶81;检测波长242 nm;流速1.0 m L·min-1,以黄体酮为内标。结果胺碘酮线性关系为y=0.227 7 x-0.000 1,r=0.999 5;去乙基胺碘酮线性关系y=0.220 6x-0.015 6,r=0.999 8,回收率>90%、RSD<10%。结论此方法简便、准确,可操作性强,可快速准确地检测出患者的胺碘酮及去乙基胺碘酮血药浓度,值得推广使用。
Objective To establish a HPLC method for the determination of plasma concentrations of amiodarone and its metabolite deethionate in patients. Methods The column of Ultimate C18 (250 mm × 4.6 mm, 5 μm) was used. The mobile phase was acetic acid-triethylamine buffer solution: methanol = 19:81. The detection wavelength was 242 nm. The flow rate was 1.0 m L · min- Ketone is an internal standard. Results The linear relationship of amiodarone was y = 0.227 7 x-0.000 1, r = 0.999 5; the linear relation of ethidium iodide was y = 0.220 6x-0.015 6, r = 0.999 8, the recovery was> 90% 10%. Conclusions This method is simple, accurate and easy to operate. It can quickly and accurately detect the plasma concentration of amiodarone and deethionate in patients and is worth popularizing.
