目的:探讨不同土壤施氮量对龙胆药材中龙胆苦苷等3种活性成分含量的影响,为提高龙胆栽培技术提供实验依据。方法:将龙胆随机分为5组,每组土壤氮含量依次升高,栽培15 d后,采用高效液相色谱法对药材龙胆中活性成分的含量进行分析,色谱柱为Waters C18(5μm,4.6 mm×250 mm),保护柱:Securityguard-C_(18)(5μm,4.0 mm×2.0 mm);流动相:甲醇-水(25∶75);流速:1 min·m L-1,柱温:30℃;进样体积:10μL;检测波长:254 nm。结果:龙胆活性成分中龙胆苦苷与土壤中含氮量成倒数相关,獐牙菜苷与土壤中含氮量成S相关,獐牙菜苦苷与土壤中含氮量无相关性。结论:一定程度内追加氮肥,能显著增加作物产量,但过量追施氮肥,也会造成作物减产。
OBJECTIVE: To investigate the effects of different amount of soil nitrogen on the contents of three active ingredients of gentiopicroside in Gentiana scabra, and to provide experimental evidence for the improvement of gentian cultivation techniques. Methods: The gentian were randomly divided into five groups. The contents of nitrogen in each soil soil were increased in turn. After cultivation for 15 days, the contents of active components in the gentian were analyzed by HPLC. The chromatographic column was Waters C18 , 4.6 mm × 250 mm), guard column: Securityguard-C 18 (5 μm, 4.0 mm × 2.0 mm); mobile phase: methanol- water Temperature: 30 ℃; injection volume: 10μL; detection wavelength: 254 nm. Results: The gentiopicroside in gentian active ingredient was inversely related to the nitrogen content in the soil. The content of urinary nitrogen in soil was related to swertia glycoside, and the content of swertia glycoside in soil was not correlated with nitrogen content in soil. Conclusion: Adding nitrogenous fertilizer to a certain extent can significantly increase the crop yield, but excessive nitrogenous fertilizer may also result in crop yield reduction.
