通过荧光光谱分析,研究了芹菜素与犬细小病毒衣壳蛋白之间的相互作用。研究结果表明,芹菜素对犬细小病毒衣壳蛋白的荧光强度具有明显的猝灭作用。获取了在293 K、298 K和303 K下的猝灭常数,证明了二者之间的作用类型为静态猝灭机制,且二者的结合位点数为1。通过求得热力学参数,证实了二者之间的作用力形式为疏水作用。由荧光响应能量转移技术求得二者之间的结合距离,并运用同步荧光光谱分析和紫外吸收光谱分析探讨了芹菜素对犬细小病毒衣壳蛋白构象的影响。
The interaction between apigenin and canine parvovirus capsid protein was studied by fluorescence spectroscopy. The results show that apigenin has obvious quenching effect on the fluorescence intensity of canine parvovirus capsid protein. The quenching constants at 293 K, 298 K and 303 K were obtained, and the interaction between them was found to be static quenching. The number of binding sites was 1. By calculating the thermodynamic parameters, it is confirmed that the interaction between them is hydrophobic. The binding distance between the two proteins was determined by fluorescence energy transfer (FTIR) technique. The effect of apigenin on the conformation of the canine parvovirus capsid protein was explored by simultaneous fluorescence spectroscopy and ultraviolet absorption spectroscopy.
