目的:通过醋酸致大鼠溃疡性模型试验,探讨久泻宁颗粒治疗急性溃疡性结肠炎的机制。方法:除空白组外,其他各组以2,4-二硝基氯苯和醋酸联合应用建立大鼠溃疡性结肠炎模型。造模成功后,将70只SD大鼠分层随机分空白组、UC模型对照组、补脾益肠丸阳性对照组、柳氮磺胺吡啶片阳性对照组、久泻宁颗粒高剂量组、中剂量组、低剂量组,每组10只(雌雄各半)。空白组与UC模型对照组给饮水,其他分别给相应的药物,连续14天后,腹主动脉采血测定:血清IL-2、TNF-α、免疫球蛋白Ig G、Ig M以及SOD、MDA,同时取结肠溃疡明显处作病理分析。结果:UC模型对照组与正常组比较,UC模型对照组明显出现免疫异常(P<0.01),血清TNF-α、Ig G、Ig M水平明显升高(P<0.01),IL-2水平明显降低(P<0.01)。与模型对照组比较,久泻宁颗粒组TNF-α、Ig G、Ig M、MDA水平降低(P<0.05~P<0.01),IL-2、SOD水平升高(P<0.05~P<0.01)。结论:久泻宁颗粒能明显调节UC大鼠机体免疫功能异常,对模型大鼠结肠溃疡面有较好的修复和恢复作用。
OBJECTIVE: To investigate the mechanism of Jiuxiaining granule in the treatment of acute ulcerative colitis by acetic acid-induced rat model of ulceration. Methods: In addition to the blank group, the other groups with 2,4-dinitrochlorobenzene and acetic acid combined application of rat ulcerative colitis model. After successful modeling, 70 SD rats were randomly divided into blank control group, UC model control group, Bupi Yichang Pills positive control group, sulfasalazine positive control group, Jiuxianning high-dose group, Dose group, low dose group, each group 10 (male and female half). The blank group and the UC model control group were given drinking water, and the others were given the corresponding drugs. After 14 consecutive days, blood samples were collected for determination of serum IL-2, TNF-α, immunoglobulin Ig G, Ig M and SOD, MDA Colon ulcer was taken for pathological analysis. Results: Compared with the normal control group, the UC model control group showed obvious immunological abnormalities (P <0.01), and the levels of serum TNF-α, Ig G and Ig M were significantly increased (P <0.01) Decreased (P <0.01). Compared with the model control group, the levels of TNF-α, Ig G, Ig M and MDA in Jiuxianning Granule group were significantly decreased (P <0.05 ~ P <0.01) ). Conclusion: Jiu-Ni-ning Granules can obviously regulate the immune function of UC rats, and have a good repair and recovery effect on the colon ulcer of the model rats.
