Due to the lack of methods for assessing the immunogenicity of oral vaccines, the efficacy and appropriate immunization dose of each new vaccine candidate must be examined through large-scale field trials. Here we report a method for determining the immunogenicity of oral vaccines by counting the number of antibody-secreting cells in the serum after immunization. The principle is: After oral immunization, lymphocytes in the small intestine lymphoid tissue are sensitized and migrate to mature local lymph nodes and then return to the intestinal mucosa through the blood circulation. This lymphocyte can exist in peripheral blood and its synthetic antibody can be measured in vitro. The authors applied this method to determine the local immune response of Ty21a oral vaccine. First, divide 80 volunteers
