目的探讨角质细胞生长因子-2(KGF-2)对角膜激光烧伤的治疗作用及其作用机制。方法采用日本大耳白兔构建激光角膜烧伤模型,实验分烧伤对照组、bFGF治疗对照组、KGF-2 12.5μg/ml组、KGF-225μg/ml组、KGF-2 50μg/ml组。通过裂隙灯显微镜与组织病理学观察、烧伤斑图像分析、角膜OD值测定和MTT法检测损伤后角膜的恢复情况,比较治疗后7、14 d各组的差异。结果 KGF-2治疗后兔角膜上皮细胞修复良好,基质中纤维板层排列整齐,无明显的炎症反应,新生血管形成较少,角膜透光率增加,损伤斑面积明显缩小,且KGF-2浓度为25μg/ml时达到最大修复。KGF-2对角膜上皮细胞无明显的促增殖活性。结论 KGF-2可通过减轻角膜激光烧伤后的各种炎症反应、加速角膜上皮细胞的再生和迁移以及促进角膜基质纤维修复等机制,促进激光烧伤后受损角膜的修复。
Objective To investigate the therapeutic effect of keratinocyte growth factor-2 (KGF-2) on corneal laser burn and its mechanism. Methods The laser keratoplasty model was established in Japanese white rabbits. The rats in burn group, bFGF treatment group, KGF-2 12.5μg / ml group, KGF-225μg / ml group and KGF-2 50μg / ml group were included in this study. Slit lamp microscopy and histopathological observation, burn plaque image analysis, corneal OD value determination and MTT method were used to detect corneal recovery after injury, and the differences between groups after 7 and 14 days of treatment were compared. Results After corneal epithelial cells were repaired by KGF-2, the corneal epithelial cells were well repaired. The fibrinous layer in the stroma was arranged neatly with no inflammatory reaction. The formation of neovascularization was less, the corneal light transmittance was increased, the lesion area was significantly reduced, and KGF-2 concentration was 25μg / ml to achieve maximum repair. KGF-2 on the corneal epithelial cells showed no significant proliferative activity. Conclusion KGF-2 can promote the repair of damaged cornea after laser burns by reducing various inflammatory reactions after corneal laser burn, accelerating the regeneration and migration of corneal epithelial cells, and promoting the repair of corneal stroma fibrosis.
