目的:观察聚腺苷二磷酸核糖聚合酶(PARP)及核因子κB(NF-κB)在重症急性胰腺炎(SAP)大鼠腺垂体中的表达及对腺垂体损伤的作用。方法:雄性Wistar大鼠40只,随机分为五组:假手术组(SO组)及SAP1、3、6、12h组。胆胰管逆行注射5%牛磺胆酸钠制备SAP模型。测定腹水量、血清淀粉酶、脂肪酶水平;光镜观察胰腺及垂体病理改变;电镜观察垂体超微结构改变;免疫组化观察垂体PARP及NF-κB的表达。结果:SAP组血清淀粉酶、脂肪酶水平及胰腺病理学评分逐渐增加,均较SO组明显升高(P<0.05)。
Objective: To observe the expression of PARP and NF-κB in the adenohypophysis of rats with severe acute pancreatitis (SAP) and its effect on the injury of pituitary gland. Methods: Forty male Wistar rats were randomly divided into five groups: sham operation group (SO group) and SAP1,3,6,12h group. The biliary and pancreatic duct retrograde injection of 5% sodium taurocholate SAP model. The levels of ascites, serum amylase and lipase were measured. The pathological changes of pancreas and pituitary were observed under light microscope. The ultrastructure of pituitary gland was observed by electron microscope. The expression of PARP and NF-κB was observed by immunohistochemistry. Results: The levels of serum amylase, lipase and pancreatic pathology increased gradually in SAP group, which were significantly higher than those in SO group (P <0.05).
