目的观察Tipe2过表达对小细胞肺癌细胞H446凋亡相关分子的影响。方法以脂质体转染技术建立Tipe2过表达的H446小细胞肺癌细胞系,利用荧光显微拍照技术和Western Blot免疫印迹技术分别检测Tipe2过表达情况和细胞凋亡相关分子Bax和Mcl-1的变化。结果 GFP-Tipe2质粒脂质体转染H446细胞高表达Tipe2,并带有标签GFP的绿色荧光。Tipe2过表达的H446细胞促凋亡分子Bax表达增高,而抗凋亡分子Mcl-1表达降低,细胞凋亡明显。结论 Tipe2过表达通过上调Bax和下调Mcl-1促进小细胞肺癌H446凋亡。
Objective To observe the effect of Tipe2 overexpression on the apoptosis-related molecules of small cell lung cancer cell line H446. Methods Liposome-mediated transfection technique was used to establish the H446 small cell lung cancer cell line overexpressed by Tipe2. The expression of Tipe2 and the expression of apoptosis related molecules Bax and Mcl-1 were detected by fluorescence microscopy and Western Blot. Variety. Results GFP-Tipe2 plasmid liposome transfected H446 cells highly expressed Tipe2 with green fluorescence of labeled GFP. The expression of Bax was increased in Tipe2 overexpressing H446 cells, while the expression of anti-apoptotic molecules Mcl-1 was down-regulated. Conclusion Tipe2 overexpression promotes apoptosis of small cell lung cancer H446 by up-regulating Bax and down-regulating Mcl-1.
