Establish a rapid, highly sensitive method for detecting Ki-ras mutations and apply them to the detection of colorectal cancer. The sensitivity of PCR-PIREMA (Primer Introduced Restriction with En-richment of Mutant AleleAssay) was 10-5 to 10-6, and 8 Ki-ras genes were detected in 30 colorectal cancer samples. In the sample with 12 codon mutations, the mutation rate was 26.7%. The RCR-PIREMA method is proved to be a rapid and highly sensitive method suitable for detecting rare mutations in a large number of wild-type copies.
