本研究旨在通过体内和体外实验探索花椒的石油醚、乙酸乙酯和正丁醇萃取物调节固醇类物质和脂多糖诱导的胆固醇积聚。通过测定总酚含量萃取物和DPPH清除率评价萃取物的抗氧化活性。在体外我们用25-羟胆固醇+胆固醇诱导HepG2细胞, 经PE、EtOAc、BuOH 预处理之后, 发现细胞总胆固醇含量和ApoB的分泌降低, SREBP2、HMGCR和ACAT的表达下调的同时CYP27A1、ABCA1和LDLR的表达上调。在体内我们用腹腔注射LPS诱导C57BL/6小鼠产生炎症, 血清TNF-α水平和促炎因子mRNA的检测结果发现EtOAc和BuOH组分有抗炎的作用。另外, BuOH萃取物能降低肝脏总胆固醇含量, 提高LXR-α、ABCA1的mRNA表达。通过活性追踪分离, 我们从EtOAc和BuOH两个组分中分离得到β-谷甾醇、芝麻素、桉脂素和syringaresinol-β-D-glucoside 4个化合物。
The aim of this study was to investigate the cholesterol accumulation induced by sterols and lipopolysaccharides in petroleum jelly, ethyl acetate and n-butanol extracts by in vitro and in vivo experiments. The antioxidant activity of the extract was evaluated by measuring the total phenolic content extract and the DPPH clearance rate. In vitro, HepG2 cells were induced by 25-hydroxycholesterol + cholesterol. Pretreated with PE, EtOAc and BuOH, the levels of total cholesterol and ApoB were decreased, and the expressions of SREBP2, HMGCR and ACAT were down-regulated while CYP27A1, ABCA1 and LDLR The increase of expression. In vivo, we induced inflammation in C57BL / 6 mice by intraperitoneal injection of LPS, and serum levels of TNF-α and proinflammatory cytokines mRNA showed that EtOAc and BuOH components have anti-inflammatory effects. In addition, BuOH extract can reduce the total cholesterol content of the liver and increase the mRNA expression of LXR-α and ABCA1. Through activity-tracing separation, we isolated 4 compounds of β-sitosterol, sesamin, eucalyptol and syringaresinol-β-D-glucoside from EtOAc and BuOH.
