目的:建立扛板归中总黄酮和槲皮素的含量测定方法。方法:采用紫外-可见分光光度法测定药材中总黄酮含量;RP-HPLC测定药材中槲皮素含量。结果:分光光度法测定总黄酮时,在0.00864~0.06912mg·mL-1的浓度范围内线性关系良好(r=0.9998),平均回收率=98.6%。RSD=1.68%;RP-HPLC测定槲皮素时,在0.008~0.04mg·mL-1的浓度范围内线性关系良好(r=0.9998),平均回收率为98.5%,RSD=1.31%。结论:所用方法准确、简便,可用于扛板归药材的质量控制。
Objective: To establish a method for determination of total flavonoids and quercetin in Pleurotus ostreatus. Methods: The content of total flavonoids in medicinal materials was determined by UV-Vis spectrophotometry. The content of quercetin in medicinal materials was determined by RP-HPLC. Results: When the total flavonoids were determined by spectrophotometry, the linearity was good (r = 0.9998) in the range of 0.00864 ~ 0.06912 mg · mL-1, the average recovery was 98.6%. RSD = 1.68%. When quercetin was determined by RP-HPLC, the linearity was good (r = 0.9998) in the range of 0.008-0.04 mg · mL-1 with the average recovery of 98.5% and RSD = 1.31%. Conclusion: The method used is accurate and simple, and it can be used for the quality control of the plates.
