建立一种高效液相色谱-蒸发光散射检测(HPLC-ESLD)同时对不同品种葡萄干中的果糖、葡萄糖、蔗糖和麦芽糖进行测定的方法。采用氨基柱(200 mm×4.6 mm,5μm)进行分离,V(乙腈)∶V(水)=75∶25为流动相,流速为1 mL/min,ELSD漂移管温度为85℃。结果标明,在该条件下4种糖的组分都能够得到较好的分离,线性良好,该方法相对标准偏差(RSDs)1.4%~3.6%,平均回收率94%~99%,4种组分的线性相关系数r>0.999 0,四种糖类在葡萄干中的方法检出限(S/N=3)为5~11 mg/kg。测定了新疆8种不同品种葡萄干中果糖、葡萄糖、蔗糖和麦芽糖的含量。所建立的方法简便、快速、稳定,重现性好。
A method for the simultaneous determination of fructose, glucose, sucrose and maltose in different varieties of raisins by high performance liquid chromatography-evaporative light scattering detection (HPLC-ESLD) was established. The separation was carried out on an amino column (200 mm × 4.6 mm, 5 μm) using V (acetonitrile): V (water) = 75:25 as mobile phase at a flow rate of 1 mL / min and ELSD drift tube temperature of 85 ° C. The results showed that all four kinds of sugars could be separated well with good linearity.
