目的 :探讨晚期糖化终产物 (AGEs)对离体新生牛胸主动脉血管平滑肌细胞 (VSMC)的影响及机制。 方法 :观察VSMC受 AGEs刺激后其生长、超微结构和血小板源生长因子 - A(PDGF- A)的变化以及维生素 E对上述变化的影响。结果 :AGEs促进 VSMC生长 ,细胞内 AGEs和 PDGF- A的免疫反应呈阳性 ,线粒体和粗面内质网增多。维生素 E不阻断 VSMC对AGEs的摄取 ,但能抑制 AGEs的细胞效应。 结论 :AGEs对 VSMC有直接效应 ,氧化应激可能参与了 AGEs的细胞效应。
Objective: To investigate the effect of advanced glycation end products (AGEs) on isolated rat aortic vascular smooth muscle cells (VSMCs) and its mechanism. Methods: The changes of growth, ultrastructure and PDGF - A of VSMC stimulated by AGEs were observed and the effect of vitamin E on these changes. Results: AGEs promoted the growth of VSMCs. The immune responses of AGEs and PDGF-A in cells were positive, and mitochondria and rough endoplasmic reticulum increased. Vitamin E does not block the uptake of AGEs by VSMC but inhibits the cellular effects of AGEs. Conclusion: AGEs have a direct effect on VSMC, and oxidative stress may be involved in the cellular effects of AGEs.
